    This paper provides a novel method for conducting ELISA in order to determine the relative stoichiometry and affinity of von Willebrand Factor (vWF) binding to coagulation factor VIII (FVIII) with the intention of creating a simple home test of vWF activity in patients with von Willebrand Disease type 2N.

    While other papers (cited by the authors) have shown novel methods for testing the relationships between vWF and FVIII, the authors provide a simple method, which can be used for testing in research laboratories.

    The authors provide adequate literature to support their claim that their method is simpler/more economically sound.

    The authors do not provide comparisons with other ELISA tests for vWF/FVIII interactions and it is therefore not possible to determine the validity of their claim that their method is easier. Nor does it provide a compairson of the accuracy of their method with other methods.

    The mothodology described in the paper is thoroughly explained. However, there was no assessment of the removal of FVIII from the vWF and it is therefore difficult to assess the accuracy of the method based on the provided information.


  • Would any other experiments or additional information improve the paper? How much better would the paper be if this extra work was done, and how difficult would such work be to do, or to provide?

    A potential test for the presence of FVIII after washing Wilate(r) might have been to conduct another ELISA on the pretreated vWF/FVIII after FVIII removal to look for the presence of FVIII. Studies comparing existing ELISA tests with the one presented in this paper, may help support the validity of the method, as well as the author's claim that their method is 'easier' to conduct.
